Data from the phase 2/3 KEYNOTE-483 study showed that pembrolizumab plus chemotherapy significantly improved overall survival vs chemotherapy alone, with a median OS of 17.3 months and 16.1 months, respectively. Moreover, the median progression-free survival was 7.1 months in both the investigative and control arms, respectively; this translated to a 20% reduction in the risk of disease progression or death. The confirmed objective response rate was also higher with the addition of pembrolizumab to chemotherapy vs chemotherapy alone, at 52% and 29%, respectively. The median duration of response in the respective arms was 6.9 months and 6.8 months. The study enrolled patients with unresectable advanced or metastatic malignant pleural mesothelioma. Patients needed to have measurable disease by modified RECIST 1.1 criteria, an ECOG performance status of 0 or 1, and acceptable hematological, liver, and renal function.2 They could not have previously received systemic therapy in the advanced setting, a contraindication to immunotherapy, no other active malignancy, and no known central nervous system metastases unless treated and stable. Study participants were randomly assigned 1:1 to receive pembrolizumab every 3 weeks for up to 2 years plus pemetrexed and platinum-based chemotherapy for up to 6 cycles or pemetrexed and platinum-based chemotherapy for up to 6 cycles.1,2 Chemotherapy consisted of cisplatin at 75 mg/m2 or carboplatin at area under the curve 5 or 6 and pemetrexed at 500 mg/m2. Patients were stratified by histology. The main efficacy outcome measure was OS and additional efficacy measures included PFS, ORR, and DOR per blinded independent central review and according to modified RECIST 1.1 criteria for mesothelioma."Reference Link

The study will enroll 40 participants who will be randomized in a 1:3 ratio to receive either a placebo or VELDONA in a double-blind trial. Participants will take VELDONA or a placebo sublingually every day for 24 weeks. The primary objective of the study is to evaluate the reduction in oral warts' surface area. A successful response is defined as a reduction of 75% or more in the surface area of oral warts compared to baseline, while a non-responsive result indicates less than 75% reduction. The secondary objectives include evaluating oral wart surface area reduction by 50% or more, as well as participant self-assessments through questionnaires comparing oral health to baseline conditions. The study's Principal Investigator will also objectively assess the participants' oral conditions. This clinical study will be conducted in accordance with Good Clinical Practice guidelines by Bestat Pharmaservices Corporation, a TFDA-certified Contract Research Organization. The study is expected to begin in November 2024, with the first patient visit anticipated in November. Patient enrollment is projected to take one year, with the last patient visit expected in May 2026, and study completion anticipated in July 2026. Protocol of the study is currently being reviewed for Investigational Review Board approval by National Taiwan University Hospital and Taiwan Food and Drug Administration. Ainos has conducted one pilot study and two phase 2 trials in the U.S. involving 77 HIV-positive patients with multiple oral warts. In these trials, patients were randomized to receive either VELDONA or a placebo, taking the medication three times daily for 24 weeks. After treatment, patients receiving VELDONA showed significant reductions in oral wart surface area, with decreases of over 75%, and in some cases up to 99%.

These reductions in reactive astrocytes corresponded with enhanced cognitive performance in the same dose groups, further indicating that aprepitant's therapeutic benefit may be tied to its impact on astrocytic activity. Reactive astrocytes, which are typically larger than normal astrocytes, play a role in the progression of neurodegenerative diseases like Alzheimer's. Hoth Therapeutics is currently conducting a secondary analysis to determine whether the size or total number of astrocytes is changing as a result of treatment. Early observations suggest that the vehicle-treated Alzheimer's mice exhibit a larger population of reactive astrocytes, while the HT-ALZ treated groups show fewer reactive astrocytes, pointing to a potential breakthrough in the treatment of Alzheimer's. Hoth Therapeutics plans to continue advancing HT-ALZ through further studies and clinical trials, aiming to provide a novel therapeutic option for patients suffering from Alzheimer's disease.

Snai generated 100% regulated revenue of EUR 947M (which is after the deduction of gaming duties) and Adjusted EBITDA of EUR 256m in financial year 2023, of which 50% was generated online.
