Tire production at Debica resumed on August 22, 2023 at approximately 55% of capacity. The facility is currently operating at approximately 70% of capacity and a full ramp-up is not expected until the fourth quarter of 2024 due to the lead time to replace the damaged equipment. "We are mitigating the impact on our customers by leveraging existing inventory and by increasing production at other facilities, which should help to offset lost production until Debica returns to full production," the company said. "Europe, Middle East and Africa consumer sales are expected to be negatively impacted by $20M to $25M in the third quarter of 2023 and $10M to $15M in the fourth quarter of 2023 due to the shutdown and limited production capacity at Debica. The related lost sales margin, as well as unabsorbed fixed costs and other period expenses, resulting from the shut-down and subsequent ramp-up period are expected to negatively impact EMEA's operating income by $10M to $15M in both the third quarter of 2023 and the fourth quarter of 2023. We will provide an estimated impact to 2024 after we have confirmed the restoration schedule. We maintain third-party insurance with a $15M per occurrence deductible that covers property damage, cleanup expenses and qualifying business interruption impacts. This Phase 1 study was designed with two parts: The dose-finding part with 1 patient at dose level 1 and 9 patients at dose level 2. The expansion part at the recommended phase 2 dose, or RP2D, with 12 additional patients. NBTXR3 was administered prior to radiotherapy, or RT, via an endoscopic ultrasound, or EUS-guided intratumoral injection. All patients received low-dose intensity-modulated radiation in 15 fractions, and were followed up to one year. Importantly, all patients in the study had previously received a four-month course of chemotherapy and showed no radiographic evidence of metastases at screening. The first patient at dose level 1 and subsequent 14 patients at dose level 2 had no injection complications. One patient at dose level 2 had 1 dose-limiting toxicity related to RT. As of the data cutoff, 13 patients were evaluable for efficacy. 11 patients had stable disease, or SD, 1 had progressive disease in the injected lesion, and 1 had a pathological complete response after surgery. Taken together, these results represent a 92.3% local disease control rate and a median Overall Survival of 21 months in evaluable patients. Notably, the patient who achieved pathological complete response entered the study with an unresectable tumor. The dose-finding part of the study is complete, establishing the RP2D at 42% of gross tumor volume, or GTV, and achieving the primary objective of the trial. The expansion part remains ongoing. The principal investigator concluded that these data suggest tolerable safety and promising early signs of anti-tumor efficacy. The company believes these results support the rationale for further development of NBTXR3 in pancreatic cancer.

Under the terms of the transaction, an affiliate of Pennsylvania American Water will merge with Appalachian and, subsequently, Appalachian will merge into Pennsylvania American Water. Appalachian is a private, investor-owned system serving approximately 1,450 customers in Avis Borough, Pine Creek Township, and Dunnstable Township, Clinton County, and is located near Pennsylvania American Water's existing Milton, Nittany, and Boggs water systems. "Appalachian has served our customers proudly since 1995, and we are grateful for our partnership with the community over this time," explained Frank Sargent, Jr., president of Appalachian. "We believe that this transaction with Pennsylvania American Water will ensure that our customers and employees are placed in the hands of another competent, community-minded utility partner going forward." In the study, Nanox.Al's HealthCCSng software was employed to assess Coronary Artery Calcification levels from non-gated, non-contrast chest CT scans. The software's categorizations were subjected to qualitative evaluation by two radiologists who reviewed each case. Patient categorization included low, moderate and severe with exclusions applied to individuals with certain medical histories or artifacts. Among the 326 eligible patients who participated in the study from January to July 2023, 101 out of 326 exhibited severe CAC, 88 had moderate CAC, and 137 showed low CAC. Patients with severe CAC levels were referred to specialized preventive cardiology clinics for in-depth evaluation and treatment. Those categorized with low and moderate CAC were directed to primary care physicians for further assessment and medical optimization.

The final readout of this study is expected by mid-2024. A positive readout would inform next development steps. Interim data from the intent-to-treat population being reported today show efficacy trends in PFS, demonstrating a delay in disease progression in the treatment arm of approximately 33% compared with the control arm, with the hazard ratio nearing the required value. Subgroup analyses show patients treated with a PARP inhibitor as maintenance therapy had longer PFS and OS if they were also treated with IMNN-001 compared with patients treated with NACT only. This was not a pre-specified subgroup as PARP inhibitors were approved after the OVATION 2 Study was initiated. The median PFS in the PARPi + NACT group and the PARPi + NACT + IMNN-001 group was 15.7 months and 23.7 months, respectively. The median OS in the PARPi + NACT group was 45.6 months and has not yet been reached in the PARPi + NACT + IMNN-001 group. While the data is still preliminary, the Company has concluded at this point that patients treated with a combination of NACT + PARPi + IMNN-001 appear to have the greatest benefit and should be the focus of on-going follow up. IMUNON also continues to see benefits in other secondary endpoints including an approximately 20% higher R0 tumor resection score and a doubling of the CRS 3 chemotherapy response score to approximately 30% in the treatment arm versus 14% in the control arm. Safety analyses continue to show good tolerability of IMNN-001 in this setting.

Importantly, these therapies are designed to work across multiple infectious diseases and threats, and could be stockpiled preemptively ahead of outbreaks. In this long-term open-label safety study, tenapanor showed acceptable tolerability with a safety profile consistent with that reported in the prior studies. "The T3MPO-3 results provide important data on IBSRELA's safety profile in a longer-term setting," said David Rosenbaum, Ph.D., chief development officer of Ardelyx. "We are pleased by the publication of these results in this distinguished gastroenterology journal as we continue to grow awareness of IBSRELA as another option for people with IBS-C." Patients who completed T3MPO-1 or T3MPO-2, the two Phase 3 trials of tenapanor for IBS-C, were eligible for enrollment in T3MPO-3. The T3MPO-3 trial enrolled a total of 312 patients of which 262 patients completed treatment. Patients received open-label tenapanor 50 mg twice a day for up to an additional 39 or 26 weeks. Ninety patients received greater than or equal to52 weeks of tenapanor. Treatment-emergent adverse events were reported in 117 of the 312 patients in the entire T3MPO-3 population and in 52 of the 90patients who received greater than or equal to52 weeks of tenapanor. Overall, 10.6% of patients experienced diarrhea. TEAEs led to limited discontinuations in the entire T3MPO-3 safety population, due to diarrhea, due to flatulence and abnormal GI sounds. Tenapanor was generally well tolerated with diarrhea being the most commonly reported adverse event.

The $440mm Class A-1 notes were marketed in a public asset backed securitization whereas the $275mm Class A-2 notes were privately placed. The Class A-1 and A-2 notes were both priced with a coupon of 6.60%. Strong investor demand for the Class A-1 notes resulted in a spread of 240bps and a yield of 6.78%, while also enabling the Class A-1 notes to be upsized by $100mm to $440mm. The spread of 240bps represents an improvement of 25bps from Sunrun's 2023-1 asset backed securitization in May 2023. The Class A-1 and A-2 notes represent an advance rate of approximately 67.4% of the securitization share of the aggregate discounted solar asset balance using a 6% discount rate. The Class A-1 and A-2 notes have an expected weighted average life of 6.53 years, an Anticipated Repayment Date of July 30, 2030 and a final maturity date of January 30, 2059. Similar to prior transactions, Sunrun raised an additional subordinated subsidiary-level non-recourse financing totaling $253mm, after the securitization transaction closed, which increased the cumulative advance rate obtained by Sunrun. The terms associated with the securitization and subsequent subordinated non-recourse financing, taken together with related tax equity proceeds, upfront incentives, and customer prepayments, net of transaction fees and required cash reserves, are equivalent to a cumulative advance rate that is consistent with the company's prior commentary of approximately 79% to 84% of the company's contracted Subscriber Value metric using a 6% discount rate. Sunrun obtained actual net proceeds that slightly exceeded this range in the most recent series of transactions, with the debt sizing benefiting from approximately a quarter of the assets in these financings being seasoned as they have been placed in service over 6 years ago. In connection with including these seasoned assets in the financing, Sunrun also repaid $255 million in total debt, including senior and subordinated term loans maturing in 2024 and 2029, respectively.

"It is important for the research community to have a benchmarking study comparing OGM to CMA, especially for genetic disorders, since OGM has a comparatively cost-effective workflow with simple interpretation and analysis. These findings compare favorably to historic partial response and disease control rates of 23% and 48%, respectively, reported with the current standard of care, the chemotherapy combination gemcitabine and nab-paclitaxel. Pancreatic ductal adenocarcinoma is the most common type of pancreatic cancer and is expected to be the second leading cause of cancer-related death in the U.S. by 2023.
