PG&E will safely restore power in stages as quickly as possible, with the goal of restoring most customers by end of day Thursday, based on the current weather conditions.

In an earlier Phase 2 study testing TSC in newly diagnosed glioblastoma multiforme, or GBM, brain cancer patients, an almost fourfold increase in two-year survival was seen versus historical controls in inoperable patients. The current Phase 3 INTACT trial is designed to examine this finding in a fully powered safety and efficacy registration study, which, if successful, could be the basis for U.S. FDA approval. In the INTACT trial, subjects are randomized at baseline to SOC for first-line treatment of GBM plus TSC, or to SOC alone. The SOC for GBM is temozolomide plus radiation therapy for 6 weeks, followed by 28 days of rest, then by 6 cycles of post-radiation temozolomide treatment. In a modification to the Phase 2 dosing regimen, patients in the INTACT trial will also receive high-dose TSC during the post-radiation chemotherapy phase. A 19-patient, open-label, dose-escalation lead-in portion to the INTACT trial was recently completed, sending a positive safety signal across all patients receiving TSC. In addition, six of the seven patients who received the high dose TSC treatment are still alive, with a median survival at the present time of 14.3 months. This is compared with 9.2 months for the historical standard of patients with inoperable GBM. Since six of the TSC-treated patients are still alive, median survival time is actually increasing with the passage of time, suggesting the INTACT trial may confirm or better the efficacy findings seen in the Phase 2 study. Patients' abilities to perform their daily activities as measured by Karnofsky performance scores increased from the baseline following completion of high dose treatment with TSC. Investigators have also reported instances of inoperable GBM patients treated with the higher dose TSC regimen leaving hospice or returning to work after treatment in the open-label portion of the study.

For the two co-primary endpoints, ubrogepant 50 mg and 25 mg showed statistically significant higher response rates for pain freedom at two hours, and the 50 mg dose demonstrated statistically significant higher response rates for the freedom from the most bothersome migraine-associated symptom at two hours. Additionally, ubrogepant 50 mg was statistically superior to placebo in the absence of light sensitivity and sound sensitivity. Ubrogepant studies show that both the 50 mg and 25 mg doses were well tolerated with an adverse event profile similar to placebo. The most common adverse reaction during the single attack studies was nausea. In a year-long extension study, the most common treatment-related adverse event was also nausea, with an incidence less than2 percent. Triptans are the most popular acute treatment for migraine representing 70 percent of prescriptions. However, for people living with migraine disease, approximately 20 percent of individuals have multiple cardiovascular risk factors, and because triptans restrict blood vessels, they may not be appropriate for these migraine patients. And for those who take triptans, as many as 30 percent may not sufficiently respond or may experience side effects. In the ACHIEVE II trial, approximately 11 percent of participants had moderate to severe CV risk factors, and 23 percent of participants previously had an insufficient response to triptans.

In an earlier double-blind, placebo controlled, randomized study which also evaluated bermekimab in the treatment of HS, the study's primary endpoint was met, demonstrating significant improvement of HiSCR in patients treated with bermekimab compared to control after 12 weeks of therapy. These results have been published in a leading peer reviewed medical journal, the Journal of Investigative Dermatology. The primary endpoint for the current study is the percentage of subjects achieving a Hidradenitis Suppurativa Clinical Response at week 12. Multiple secondary efficacy endpoints will be assessed after 12 and 16 weeks of therapy, including: Numerical Rating Scale for pain and itch; Modified Sartorius Score; Dermatology Life Quality Index; Hospital Anxiety and Depression Scale; and Patient Global Impression of Change and Severity.

SRK-015 is a highly selective inhibitor of the precursor, or latent form, of myostatin, and was specifically designed to avoid interactions with related targets such as activins, GDF-11, or BMPs, to potentially improve the therapeutic profile compared to traditional non-selective inhibitors. The Phase 2 proof-of-concept trial is evaluating the safety and efficacy of SRK-015 dosed intravenously every four weeks over a 12-month treatment period. The trial is anticipated to enroll approximately 55 patients with Type 2 or Type 3 SMA in the U.S. and Europe across three distinct cohorts. Patients in Cohorts 1 and 2 are being treated with 20 mg/kg of SRK-015 Q4W and patients in Cohort 3 are randomized to either 20 mg/kg or 2 mg/kg Q4W. The primary objectives of the cohorts are to assess safety and clinically meaningful motor functional outcomes, such as the Revised Hammersmith Scale and the Hammersmith Functional Motor Scale Expanded. The preliminary PK/PD analysis of the TOPAZ trial includes data from 29 patients across the three cohorts; 12 patients in Cohort 1, eight patients in Cohort 2, and nine patients in Cohort 3. These patients had received one dose of SRK-015 and were evaluated for four weeks as of the data cutoff. The preliminary results are as follows: Dose-dependent increases of up to 100-fold in serum latent myostatin levels following treatment with SRK-015 confirms the presence of latent myostatin in patients with SMA and demonstrates robust target engagement. Fold-increases from baseline in serum latent myostatin levels in the first four weeks following SRK-015 treatment were comparable between SMA patients in the TOPAZ trial and healthy adult volunteers in the Phase 1 trial. In patients with SMA, SRK-015 displayed a preliminary PK profile exhibiting dose proportionality and low variability, consistent with PK observations from the Phase 1 trial in healthy adult volunteers. No clinically significant safety signals had been observed as of the data cutoff for this preliminary PK/PD analysis. An interim efficacy and safety analysis is planned, encompassing a subset of patients with at least six months of treatment exposure. The interim results are expected in the first half of 2020 with top-line results for the full 12-month treatment period expected starting in the fourth quarter of 2020 and through the first quarter of 2021.

For the study to be successful, the lower bound of the 95% confidence interval of the response rate had to be at least 90%. Five key secondary endpoints demonstrated superiority to leuprolide acetate, including rapid suppression of testosterone at Day 4 and Day 15, profound suppression of testosterone at Day 15, rapid suppression of prostate-specific antigen at Day 15, and suppression of follicle-stimulating hormone at Week 24. In addition, relugolix demonstrated non-inferiority to leuprolide acetate on sustained testosterone suppression through 48 weeks with a between-group difference of 7.9%, the primary endpoint required for regulatory submissions outside of the U.S. In addition, the pharmacodynamic results showed no testosterone flare after initiation of relugolix and mean testosterone levels returned to normal levels within 90 days after treatment discontinuation. The overall incidence of adverse events in the relugolix and leuprolide acetate groups was comparable. In the relugolix group, 3.5% of men discontinued the study early due to adverse events compared with 2.6% of men in the leuprolide acetate group. The most frequently reported adverse events, reported in at least 10% of men in the relugolix group, were hot flashes, fatigue, constipation, diarrhea, and arthralgia. Unadjudicated major adverse cardiovascular events were reported in 2.9% of men in the relugolix group versus 6.2% of men in the leuprolide acetate group. These events included non-fatal myocardial infarction, non-fatal stroke, and all-cause mortality.

The patient with TDT has the beta0/IVS-I-110 genotype and required 16.5 transfusions per year before enrolling in the clinical study. The patient achieved neutrophil engraftment 33 days after CTX001 infusion and platelet engraftment 37 days after infusion. Two serious adverse events occurred, neither of which the principal investigator considered related to CTX001: pneumonia in the presence of neutropenia and veno-occlusive liver disease attributed to busulfan conditioning; both subsequently resolved. At nine months after CTX001 infusion, the patient was transfusion independent and had total hemoglobin levels of 11.9 g/dL, 10.1 g/dL fetal hemoglobin, and 99.8% F-cells. The patient with SCD experienced seven vaso-occlusive crises per year before enrolling in the clinical study. The patient achieved neutrophil and platelet engraftment 30 days after CTX001 infusion. Three SAEs occurred, none of which the PI considered related to CTX001: sepsis in the presence of neutropenia, cholelithiasis, and abdominal pain, all of which resolved. At four months after CTX001 infusion, the patient was free of VOCs and had total hemoglobin levels of 11.3 g/dL, 46.6% fetal hemoglobin, and 94.7% F-cells.

The remaining $5.575M in cash owed represents a secured obligation of Tonogold with monthly payments starting in November 2019, through June 2020. Once paid, Tonogold will own 100% of Comstock Mining, the owner and operator of the permitted Lucerne mine. Comstock and Tonogold also executed a Lease-Option agreement to use the Company's American Flat processing facilities for processing mineralized materials from the Lucerne mine. To maintain the option, Tonogold will reimburse the Company for all expenses related to owning and maintaining the facility.
