Key elements of MediciNova's Phase 3 clinical trial plan for MN-166 in progressive MS include the following: MediciNova's Phase 3 trial will enroll only subjects with secondary progressive MS without relapses. The rationale for enrolling this type of progressive MS is the following: The FDA agreed that a population of subjects with secondary progressive MS without relapses is an appropriate target population. MediciNova believes that subjects with secondary progressive MS without relapses will have the best clinical response to MN-166 treatment, as compared to other types of progressive MS. As MediciNova previously reported in April 2019, results of the subgroup analysis of the SPRINT-MS Phase 2b trial of MN-166 in progressive MS showed that the trends for reduction in the risk of confirmed disability progression, as measured by EDSS, were highest for the subgroup of subjects with secondary progressive MS without relapses, in which MN-166 demonstrated a 46% risk reduction compared to placebo as indicated by the hazard ratio of 0.538. MediciNova believes that the unmet medical need is highest in subjects with secondary progressive MS without relapses as compared to other types of progressive MS. Unlike primary progressive MS and relapsing secondary progressive MS, there are no drugs approved for long-term treatment of secondary progressive MS without relapses. In addition, secondary progressive MS without relapses is the largest subgroup of progressive MS patients. Based on the data from recently completed clinical trials in progressive MS, MediciNova believes that more than 80% of patients with secondary progressive MS do not have relapses. The FDA agreed that the primary endpoint of the Phase 3 trial should be time to 3-month confirmed disability progression, as measured by EDSS. This is the same primary endpoint that was used in the Phase 3 trials of other drugs recently approved for progressive MS. MediciNova plans to conduct a single Phase 3 trial. The FDA acknowledged that a single trial can be the basis for marketing approval and the acceptability of a single trial to support drug approval depends on the study results. The two drugs approved for relapsing secondary progressive MS in March 2019 were approved by the FDA after conducting a single Phase 3 trial.

16:41 EDT Global PC shipments grew 1.5% in Q2, Gartner says - After two quarters of decline, the worldwide PC market grew 1.5% in the second quarter of 2019, according to preliminary results by Gartner, Inc. Shipments totaled 63 million units in the second quarter of 2019, up from 62 million units in the second quarter of 2018. "Worldwide PC shipments growth was driven by demand from the Windows 10 refresh in the business market in the second quarter of 2019. Desktop PC growth was strong, which offset a decline in mobile PC shipments," said Mikako Kitagawa, senior principal analyst at Gartner. The ongoing trade dispute - and potential imposition of tariffs - adds uncertainty to the near-term outlook for PC demand. "While the U.S.-China trade war did not impact the PC market in the second quarter of 2019, the next phase of tariffs could have significant impact. Most laptops and tablets are currently manufactured in China and sales of these devices in the U.S. could face significant price increases if the punitive tariffs are imposed and vendors do not take quick action to respond," said Kitagawa. The top three vendors - Lenovo (LNVGY), HP Inc. (HPQ) and Dell (DELL) - accounted for 64.1% of global PC shipments in the second quarter of 2019, compared with 60.7% of shipments in the second quarter of 2018. These top three vendors continued to gain share in the PC market taking advantage of economies of scale. Intel's (INTC) CPU supply shortage in the first half of the year accelerated this trend. Other companies in the PC space include Apple (AAPL), Micron (MU), Western Digital (WDC), and Seagate (STX).

The legislation will now move to Governor's Office for consideration. Governor Gavin Newsom has 12 days to either sign the measure or veto it. Cadiz issued a statement regarding the Assembly vote: "As our Governor has repeatedly emphasized, California continues to suffer from chronic water supply and affordable housing shortages. These problems are not unrelated. Solutions to these two critical problems depend on infrastructure improvements that are opposed at every turn. Bills like SB 307 will make solutions even tougher to achieve. An earlier version of the Cadiz Water Project was approved by the United States Department of the Interior in 2002. A revised Project underwent a decade of additional study, peer review, state environmental impact evaluation, a separate approval by San Bernardino County and was repeatedly validated by the California Courts. SB 307 is based on the false premise that the federal government never reviewed the Project and that CEQA, the County and the Courts were not fair and independent reviewers. Opponents have parlayed their considerable political influence to establish a new state review in a new forum sought by opponents after failing to succeed in all other venues. Joined by dozens of state and local organizations, we have expressed concern because SB 307 encourages infrastructure opponents to politicize project review and singles out one California Company and one project for unique treatment. We are also supportive of the announced efforts of Three Valleys Municipal Water District to initiate an inclusive study plan over coming months to further study mountain springs in the Project area. Any application for the finding required by SB 307 as a precondition to wheeling water through the Colorado River Aqueduct will be accompanied by clear and unequivocal data and reports that can withstand critical scrutiny. We will continue to go about our business of efficiently and sustainably farming our land, acting as responsible private stewards of a public resource. The Project will be lawfully progressed under all federal, state and local regulations. More than 10 billion gallons a year are wasted to the atmosphere each year from the Bristol and Cadiz Dry-Lakes. The Project would conserve this water and divert it to 400,000 people a year. More than 63 billion gallons have been irretrievably lost since the Project was first approved. If California is serious about meeting its commitment to provide, clean, reliable and affordable water to all people, of all color in all locations, Cadiz is ready to play its part."

However, HOOKIPA's approach is designed to directly reprogram the patient's immune system with an off the shelf systemic administration. This approach eliminates the need for any ex vivo logistics that burden adoptive cell therapy and any related ex vivo cellular approaches. HOOKIPA intends to start the Phase 1/2 clinical trial in the second half of 2019 with preliminary safety and efficacy data expected in late 2020 or early 2021. It will be HOOKIPA's first clinical trial in immuno-oncology. In addition to obtaining both safety and efficacy of the targeted patient population, this program will also have a built-in translational component which will allow HOOKIPA to validate its mechanism of action within patients and further drive clinical development via a rigorous scientific method. As a result, we now control significant market share for this injectable non-opioid metastatic cancer palliation drug in North America and much of the world. MetastronTM is approved for sale in 22 countries as a non-opioid therapy for the debilitating pain associated with metastatic skeletal cancer. As part of the Metastron acquisition, we agreed to transition manufacturing to another facility and chose IsoTherapeutics to be that manufacturer in parallel with our Strontium-89 product. 'The FDA approval of IsoTherapeutics to produce a commercial drug product is one of several milestone catalysts we expect to positively impact our business over the next few months," stated CEO Denis Corin. 'Our Strontium-89 products are our lead revenue opportunities and an important step for both Q BioMed and the many patients that will benefit from finally having access to the non-opioid palliation treatments. The studies evaluated radiation exposure to healthcare professionals who cared for patients treated with Iomab-B and the exposure from handling blood samples from patients following Iomab-B infusion. Both studies reported minimal radiation exposure to the respective healthcare professionals. The SIERRA trial is a multicenter, 150 patient study for patients with active, relapsed or refractory AML or Acute Myeloid Leukemia that is the only randomized Phase 3 trial to offer a potentially curative BMT to this patient population for which there is no standard of care. Radiation exposure levels to all staff were found to be minimal, with mean cumulative exposure of 0.09 mSv, contributing minimal exposure to the 50 mSv annual occupational limit.

In the PROMISE-2 trial for chronic migraine, impact scores as measured by the Headache Impact Test were significantly elevated at baseline. The analysis, conducted within PROMISE-2, evaluated the efficacy and related functional and health-related quality of life outcomes of eptinezumab in those patients enrolled in the trial who, during a screening, reported very often or always experiencing severe headaches per HIT-6. Efficacy endpoints that were evaluated included: change from baseline in HIT-6 total score and frequency of severe pain during headaches, as well as change from baseline in the 36-item Short-Form Health Survey which is a general measure of health-related quality of life. Highlights include: Over half of eptinezumab-treated patients experienced greater than or equal toone-category decrease in the frequency of severe headaches by Month 1, compared to 43.7% for placebo. The decrease for patients treated with eptinezumab or placebo remained relatively consistent through the six-month study period. Eptinezumab treatment resulted in clinically meaningful improvements in HIT-6 scores as early as Month 1 after treatment, which were maintained or further improved throughout the six-month study period, compared to placebo, which did not achieve a clinically meaningful improvement until Month 6. In addition, eptinezumab treatment resulted in clinically meaningful improvements greater than placebo in SF-36 scores as early as Month 1 after treatment and through the six-month study period."

The company said, "In its review, ICER fails to incorporate available data on both long-term outcomes and quality-of-life. ICER's analysis and the public meeting discussion discounted patient and community perspectives on the physical, social and psychological benefits of desensitization. The resulting final report is biased against immunotherapy, generally, and fails to specifically capture the full value of AR101. Aimmune joins advocates and clinical experts asking payers and policymakers to engage in more thoughtful, unbiased consideration of available evidence-including patient perspectives on quality-of-life improvements and relief from the stress and fear of accidental exposure-when evaluating emerging treatments for peanut allergy. In clinical trials, AR101, Aimmune's investigational biologic drug for use in oral immunotherapy, has demonstrated the ability to increase the median tolerated dose of peanut protein by 100-fold in each of its two pivotal Phase 3 clinical trials. While the report found AR101 to be cost-effective well below traditional health economic thresholds, ICER failed to acknowledge the availability of the positive long-term efficacy and quality-of-life data from the PALISADE open label follow-on study, as well as the clinical outcomes data from the European Phase 3 ARTEMIS trial. These data were recently presented at the 2019 European Academy of Allergy and Clinical Immunology (EAACI) Congress and shared with ICER in May under its "academic in confidence" policy and throughout the review process including the June 11 public meeting. ICER did not acknowledge or incorporate the existing data in its final evidence report or the associated cost-effectiveness analysis. Additionally, ICER failed to consider the potential for long-term disease modification consistent with allergen immunotherapies and mischaracterized the allergic response associated with immunotherapy as a harm, rather than an expected, well understood and easily managed part of the immunotherapy process. With all immunotherapy, allergic reactions are expected and acceptable in the short term in exchange for long-term desensitization to protect against accidental exposure. In clinical trials, median tolerated dose for AR101-treated patients improved from 10mg at baseline to 1000mg at month 12 to 2000mg at month 18-demonstrating both the potential for long-term disease modification and the potential for significant protection against the average real-world reaction-provoking exposure of 125mg, or half a peanut." The FDA's Allergenic Products Advisory Committee, or APAC, will review Aimmune's biologics license application, or BLA, for AR101 at its meeting scheduled for September 13. The FDA accepted the BLA for AR101 in March and previously informed Aimmune that completion of its review would be targeted by late January 2020.

Detailed data will be shared at the American Headache Society's 61st Annual Scientific Meeting in Philadelphia that further support eptinezumab's clinical profile, including data on migraine-free months, migraine severity and quality of life. The 100% responder rate analyses evaluated the efficacy of eptinezumab in terms of the number of months a patient experienced no migraine days, i.e., the number of migraine-free months, during the one-year study period in the PROMISE-1 trial and the six-month study period in the PROMISE-2 trial. At baseline, patients enrolled in PROMISE-1 were experiencing 8.6 mean monthly migraine days per month and patients in PROMISE-2 were experiencing 16.1 mean monthly migraine days per month. Post-hoc analyses were conducted that quantified the number of patients who experienced no migraine days during each month in the trial study period. Highlights include: PROMISE-1: 18.1% of episodic migraine patients treated with 100 mg of eptinezumab and 25.2% of those treated with 300 mg of eptinezumab experienced no migraine days for at least half of the study period compared with 12.6% of placebo-treated patients; 63.3% of episodic migraine patients treated with 100 mg of eptinezumab and 64.4% of those treated with 300 mg achieved at least one migraine-free month compared to 47.7% of those on placebo. No additional patients are needed for submission of the BLA to support approval of narsoplimab in this indication. The company must meet with the program's EMA rapporteurs, scheduled to begin this summer, to refine further the path to European approval. Omeros intends to harmonize the contents of the MAA and BLA. Omeros will also need to complete and submit the BLA and MAA. FDA has previously confirmed that Omeros may submit this BLA on a rolling basis and the company is finalizing for review by FDA its proposed schedule of module submissions under that rolling BLA. As also previously reported, the nonclinical sections of the BLA have been written and are expected to comprise the first module of the BLA submission. The acquisition is expected to close in the third quarter of 2019, subject to customary closing conditions. AFI is currently overseeing a wind-down of the facility, which is expected to continue through Fall 2019. During this time, Cronos Group will also begin aligning specifications for the equipment and manufacturing required for the production and downstream processing of cannabinoids. To support this work, Cronos Group expects a team of engineers, scientists, production and quality assurance personnel currently working at the facility to join Cronos Group. 06:50 EDT Eli Lilly announces Phase 3 Emgality study results published - Eli Lilly announced that the New England Journal of Medicine published Phase 3 study results of Emgality for the treatment of episodic cluster headache. In this study, Emgality reduced the frequency of weekly cluster headache attacks across Weeks 1 to 3 compared to placebo. The FDA approval of Emgality for the treatment of episodic cluster headache in adults in June was based on these results. The study enrolled 106 patients who were randomized 1:1 to receive either 300 mg Emgality or placebo. Prior to enrollment, patients averaged 17.8 weekly cluster headache attacks in the Emgality arm and 17.3 attacks in the placebo arm. Results showed that patients taking Emgality had an average reduction of 8.7 weekly cluster headache attacks across Weeks 1 to 3 compared to a 5.2 average reduction in attacks for placebo. Additionally, 71.4% of patients treated with Emgality had their weekly cluster headache attack frequency reduced by 50% or more at Week 3, the key secondary endpoint, compared to 52.6% of placebo-treated patients. Overall, the safety profile observed in patients with episodic cluster headache treated with Emgality 300 mg monthly is consistent with the safety profile in patients with migraine treated with Emgality 120 mg monthly. Two Emgality-treated patients discontinued double-blind treatment during the episodic cluster headache study because of adverse events.

As part of its ongoing simplification/modernization initiative, the company is optimizing its operations globally, including footprint rationalization. The company's proposal is to close its manufacturing facilities in Essen and Lichtenau as well as its distribution center in Neunkirchen, all in Germany. Additionally, the company announced its decision to close its manufacturing facility in Irwin, PA. These restructuring actions advance the company toward the fiscal 2021 financial targets announced at its most recent Investor Day. They are also part of the previously announced goal of reducing plants through simplification/modernization, with additional plants currently under evaluation. The proposed restructurings are expected to be executed over the next two years. Facility closures in fiscal 2020 are part of the current restructuring action, which in total are expected to deliver estimated annualized savings of $35M-$40M and $55M-$65M in pre-tax charges. Fiscal 2021 closures would result in further structural cost reductions with estimated annualized savings of $25M-$30M. Most of these savings will be achieved in fiscal 2021 with full run rate savings being realized in fiscal 2022. The company is expected to incur pre-tax charges of $60M-$75M through fiscal 2020 and 2021 for this restructuring. These charges are primarily cash with the majority being spent in fiscal 2021. Under the proposed measures, the company would consolidate its Essen and Lichtenau operations, including both Kennametal and WIDIA branded products, into other lower-cost Kennametal Industrial facilities. The Irwin operations would be consolidated primarily into the newly modernized Infrastructure plant in Rogers, AR. Kennametal also proposed plans to outsource the distribution operations in its Neunkirchen and Essen facilities to a third-party logistics specialist. The closure plans are contingent on the negotiations with the relevant employee representatives. The company will continue to maintain a strong presence in Germany, including planned modernization investments in other Kennametal locations in the country.

While RB has acted lawfully at all times and expressly denies all allegations that it engaged in any wrongful conduct, after careful consideration, the Board of RB determined that the agreement is in the best interests of the company and its shareholders. This is a non-criminal resolution and is on the basis that there is no admission of any violation of law or any wrongdoing by RB or any RB Group employee.The settlement amount will be funded through existing borrowing facilities and cash generation. RB has a provision of $400M and will increase its provision to $1.5B at its half-year results. RB believes this will cover the settlement and any remaining litigation exposures.
